Multifunctional nanosensors for simultaneous biocidal applications including cancer therapy have emerged as a new class of materials, providing the required safety for biomedical applications in a cost-effective manner. Polymer/metal nanosensors with a unique, tumor-targeting poly(L-lysine)-based NIR-responsive polymer, poly(n-butylcyanoacrylate-co-N-(2-acryloyloxy-2-methylpropyl)-2-dodecyl-aminoethylammonium bromide) (PBCA-PAMA), were developed. In contrast to other multifunctional nanosensors, they responded to near-infrared (NIR) light (λmax = 700 nm) via an uncomplicated, large spectral change observed by UV-vis absorption spectroscopy, triggered by a rapid lysosomal/endosomal release of peroxide ions from the PAMA-conjugated polymer. The nanosensors showed an excellent photothermal response to NIR irradiation, which allowed them to be rapidly digested by glutathione peroxidase-8 (GPx-8). The nanosensors also showed effective in vitro and in vivo anticancer efficacy. Thus, a safe, selective, and effective NIR-responsive polymer/metal-based nanosensors were developed, with the potential to be applied in cancer therapy via the effective generation of cytotoxic radicals for use in the photoacoustic imaging/photodynamic therapy.
